Cílem práce je příprava speciálních surfaktantů, které po navázání na magnetické nanočástice kobalt ferritu (CoFe2O4), předají své vlastnosti uspořádané achirální struktuře kapalných krystalů (mesofázi). Surfaktant byl navržen tak, aby ve své struktuře obsahoval chirální a fotosenzitivní složku. Zavedením do kapalně-krystalické matrice tak dojde k indukování chirálního uspořádání, zároveň se celý systém stane citlivým na UV záření a získá magnetické vlastnosti propůjčené nanočásticemi. Vzniklou chirální mesofázi pak bude možné ovlivnit mechanicky, magneticky, světlem i elektrickým polem, což je v případě uspořádaných tekutin zcela unikátní fenomén. Jako chirální stavební prvek dopantu použijeme (R)-1,1'-bi-2-naftol (BINOL). Fotosenzitivní vlastnosti zajistí azoskupina, která při absorpci UV záření reverzibilně změní konfiguraci dvojné vazby z E na Z a tím dojde k rozvolnění struktury mesofáze. Surfaktanty se pak liší v délce alkoxylových řetězců ramen obsahujících azoskupinu (Obr. 1).

Synthesis of chromone-based DC-SIGN inhibitors
DC-SIGN is an immune system receptor important in the processes of antigen presenting, but also subject to misuse by many pathogens (e.g., HIV). Exploiting the medicinal potential of DC-SIGN is challenging due to the shallow and hydrophilic nature of the receptors’ primary carbohydrate binding site. A pioneer study of secondary binding sites was performed by Rademacher et al. These secondary sites are less hydrophilic and could interact with non-carbohydrate drug-like molecules. In continuity with our previous research on quinolone derivatives as DC-SIGN inhibitors, my work focuses on the synthesis and bioactivity evaluation of chromone derivatives.
The synthetic route begins with the Vilsmeier–Haack reaction of substituted 2-hydroxyacetophenone, forming the chromene-3-carbaldehyde core. Using the Pinnick oxidation, the aldehyde is then converted to an acid. Subsequently, nucleophilic fragments with suggested DC-SIGN affinity will be coupled; from these, 1-(2-(trifluoromethyl)phenyl)piperazine has already been tried.
The affinity evaluation of the synthetized chromone-based compounds is in progress in Prof. Rademacher’s group in Vienna.

Galectins are family of 15 biologically active proteins, commonly found in human tissues. The specific inhibition of human galectins has a vast pharmacological potential. Especially the inhibition of human galectin-3 (Gal-3) is desirable because of its direct link to tumour genesis and other cancer related processes. Even though all β-galactosides are Gal-3 ligands in general, previous study has shown that Gal-3 recognise N,N'-β-diacetyllactosamine epitope with high selectivity compared to other galectins. In this work, we describe a partial synthesis of novel tetra-saccharide galectin inhibitor with di-N,N'- diacetyllactosaminide epitope. The synthesis of the target compound consists of the preparation of suitably protected monomeric units and their connection via stereoselective chemical glycosylation. The initial synthetic plan had to be modified several times due to the unexpected issues with reactivity or selectivity which led to a series of dead ends. Such dead ends however illustrated the complexity of oligosaccharide synthesis and provided valuable insight into the reactivity of synthetic intermediates going beyond current state-of-the-art.

Model for PROTAC-based targeting of influenza polymerase
Proteolysis targeting chimeras (PROTACs) provide a novel approach for specific protein degradation. This emerging field has proven its therapeutic potential over the past few years. PROTACs consist of two main parts: Protein of interest (POI)-binding and E3 ligase-binding. The mode of action is driven by the proximity effect, where the PROTAC links the POI and the E3 ligase closely together resulting in polyubiquitination of said POI leading to subsequent proteasome mediated degradation. This mechanism provides many benefits over the standard inhibition model such as catalytic properties, ability to target undruggable proteins and higher resistance to mutations. Antiviral PROTACs thereby present a different perspective for fighting viral infections. This fact led to an opportunity to develop a PROTAC-based model for targeting the influenza virus, namely its polymerase. This project focuses on the PB2 subunit responsible for cap-binding, a process crucial to viral protein expression. Herein, we developed an experimental and synthetic approach for synthesis of derivatives similar to the successful small-molecule inhibitor for PB2 (VX-787) with marginally worse activity, followed by the selection of the most suitable one for the final PROTAC and investigation of its activity.

Strain-promoted [3+2] cycloaddition of cyclic alkynes with fluorinated azides
Strain promoted azide-alkyne cycloaddition (SPAAC) is an important tool for biorthogonal chemistry – bioconjugations, bioimaging and even drug delivery. For its application, the reaction needs to be selective, fast, and most importantly biocompatible. In our group we study the preparation and reactivity of new stable α-fluorinated azides. The aim of this work is to show that α-fluorinated azides can be used as reagents in SPAAC. Their reactivity with cyclooctyne derivatives DIBAC or BCN was investigated, and the reaction rate constants were determined by IR monitoring. Moreover, the analogous nonfluorinated azide was also studied for a reactivity comparison. Lastly the impact of water, buffers, and common biomolecules, such as glutathione and lysine, on the stability of α-fluorinated azides was studied to ascertain their biocompatibility.

Syntéza ligandů pro multivalentní interakce s galektiny
Galektiny jsou molekuly patřící do skupiny lektinu, což jsou proteiny mezi jejichž vlastnosti patří schopnost vázat β-galaktosidy či oligosacharidy zakončené β-D-galaktózou. Toto navázání může následně ovlivňovat řadu biologických procesů, jako je například apoptóza či regulace imunitního systému. Galektiny také figurují v různých chorobách, jako jsou záněty, srdeční onemocnění či dokonce rakovina. Cílem práce je syntéza glykocalixarenů, kdy postranní řetězce calixarenů, nesou ligandy obsahující β-D-galaktózový motiv. Takto vytvořené multivalentní systémy by měly mít vysokou afinitu ke galektinům, a tak působit jako jejich inhibitory. V porovnání s předchozími glykocalixareny jsou tyto calixareny modifikovány dialkylaminoethylovými skupinami (R2NCH2CH2-), které zvyšují rozpustnost v biologických systémech, jak bylo ukázáno na derivátu samotného calixarenu s tímto uskupením, kdy se navíc zjistil jeho inhibiční účinek růstů nádorů hlavy a krku na myších modelech.

Hit expansion on novel antiviral compounds: investigating the replacement of a sulphonamide linker with an amine
Dengue is a mosquito-borne viral disease causing an estimated 100 million infections per year. The development of antivirals against dengue is still in its early stages. Inhibition of protein-protein interactions in viral replication complexes is a promising strategy to disrupt viral replication. NS3 and NS5 complex plays a vital role in the dengue life cycle and its structure is conserved throughout other dengue serotypes, which makes it an attractive target for broad-spectrum antivirals. Docking-based virtual screening of 1 million compounds revealed 30 possible small molecules suitable for disrupting the NS3/NS5 complex. Further in-vitro analysis showed exceptional activity of sulphonamide compound A. Substitution of the sulphonamide group with an amine should bring multiple benefits, such as shortening the synthetic route or stabilization of the compound. Moreover, it should give us clues about the role/necessity of the sulphonamidic group in the biological activity of the original hit compound. The goal is to prepare compound B via 4-step synthesis and develop analogues using different heteroaryl substituents in the highlighted position.

Syntéza a studium vlastností nové generace opticky aktivních spiropyranových fotopřepínačů
Spiropyrany jsou heterocyklické fotochromní sloučeniny schopné přecházet mezi nízkopolární spiroformou a otevřeným, zwitteriontovým merocyaninem (viz. obr.). Právě díky této dramatické změně nejen své geometrie, ale především polarity, nalezly spiropyranové fotopřepínače již řadu aplikací v moderních materiálech.
Soudobý trend ve výzkumu chytrých materiálů často zahrnuje implementaci chirality, která umožňuje vznik orientovaných domén materiálu, a tím otevírá prostor pro zcela nové, často neočekávané aplikace. Spiropyrany jsou ve své povaze látky chirální díky asymetrickému spirocentru. Avšak příprava opticky čistého spiropyranového fotopřepínače je prakticky nedosažitelná, jelikož spiropyrany podléhají spontánní racemizaci skrze otevřenou merocyaninovou formu.
V naší pracovní skupině se věnujeme přípravě derivátů spiropyranu s pomocným centrem chirality v poloze 3‘. Toto centrum řídí rovnováhu spontánního zavírání merocyaninové formy za vzniku dvou epimerů, kde je v ideálním případě vznik jednoho z nich zcela potlačen, a tím získán opticky čistý spiropyran.
V rámci této práce byly připraveny tři modelové fotopřepínače na bázi spiropyranu s centrem chirality v poloze 3‘ a prodlužujícím ramenem v poloze 6. Cílové sloučeniny byly studovány pomocí NMR a UV-Vis spektroskopie.
